Abstract

ABSTRACT Lansau Kita Tea is a traditional medicine that has been formulated from lansau herbs originated from Muna Regency, Southeast Sulawesi. This study aimed to determine the safe dose that does not cause toxic effects (No Observed Adverse Effect Level / NOAEL) from Lansau Kita tea. This study was designed using Post Test Only Controlled Group Design on 30 male mice which were randomly divided into six groups. The control group was given distilled water and the test group was given Lansau Kita tea with concentrations of 2.6 mg/kg; 5.2 mg/kg; and 10.4 mg/kg of rats’ body weight for 30 days orally. Meanwhile, the satellite group was given the tea with a dose of 10.4 mg/kg, and the satellite control group was given distilled water to see the reversibility effect. Treatment outcomes were compared based on macroscopic and microscopic observations between groups by looking for signs of toxicity. The results show that the clinical biochemical levels of AST and ALT were still in the normal range. Based on statistical analysis, the relative body weight index of the heart, liver, lungs, and spleen did not show a significant difference (p <0.05). Histopathological features of the liver and kidney show that the Lansau Kita test did not cause microscopic structural changes and the toxic effect that occurs was reversible after the oral route was stopped. Comparison of the results of macroscopic and microscopic conditions from the dose test given shows that the tea is safe and does not cause toxic effects (NOAEL).Keywords:  Lansau Kita Tea,  Toxicity Subchronic Test, NOAEL  ABSTRAKTeh Lansau Kita adalah obat tradisional yang telah diformulasikan dari ramuan lansau asal Kabupaten Muna, Sulawesi Tenggara. Penelitian ini bertujuan untuk mengetahui dosis keamanan yang tidak menimbulkan efek toksik (No No Observed Adverse Effect Level / NOAEL) dari teh Lansau Kita. Penelitian ini dirancang dengan menggunakan Post Test Only Controlled Group Design pada 30 mencit jantan yang dibagi secara acak menjadi 6 kelompok. Kelompok kontrol diberikan air suling dan kelompok uji diberikan dengan seduhan teh Lansau Kita berbagai konsentrasi 2,6mg/kgbb; 5,2 mg/kgbb; dan 10,4 mg/kgbb tikus selama 30 hari dengan rute oral. Kelompok satelit dengan dosis 10,4 mg/kgbb dan kelompok kontrol satelit diberi air suling untuk melihat efek reversibillity. Hasil pengobatan dibandingkan berdasarkan pengamatan makroskopik dan miscroscopic antara kelompok dengan melihat tanda-tanda toksisitas. Hasil penelitian menunjukkan bahwa tingkat biokimia klinis AST dan ALT menunjukkan masih dalam kisaran normal. Berdasarkan analisis statistik dari indeks bobot organ relatif jantung, hati, paru-paru dan limpa tidak menunjukkan perbedaan yang signifikan (p <0,05). Gambaran histopatologis hati dan ginjal menunjukkan bahwa uji Lansau Kita tidak menyebabkan perubahan struktural mikroskopis dan efek toksik yang terjadi reversibel setelah rute oral dihentikan. Perbandingan hasil keadaan makroskopis dan mikroskopis dari uji dosis yang diberikan aman dan tidak menimbulkan efek toksik (NOAEL).Kata kunci : teh Lansau Kita, uji toksiistas subkronik, NOAEL
